This is a Spider Silk Generating Device that uses Fos and Jun leucine zipper sequences flanking a spider silk protein hexamer. The leucine zippers are linked to the spider silk hexamer by 8 AA linkers. RBS+Leader+ATG are part of the start of the gene, and the terminator is at the end.

This is a spider silk generating device that produces a hexamer of spider silk protein codon optimized for expression in E. coli. The DNA of the device is made up of Fos  and Jun linked to the front and back of the spider silk hexamer. Fos and Jun are proteins with leucine zippers that form heterodimers in solution. The idea is that our E. coli cell will produce the hexamers of spider silk and have them aggregate together in very long chains bound together by Fos and Jun interactions. Our part was placed in a Rhamnex 67K Xbrane plasmid, meaning that the device is activated by rhamnose input.

Spider Silk Actuator
