We created the arpA-binding promoter by positioning the A-factor receptor protein(ArpA) binding sequence between and partially overlapping consensus -35 and -10 hexamers with 18 bp between the hexamers. When A-factor represents and binds to ArpA proteins, the protein structure of ArpA changes and ArpA has less affinity to its DNA binding site, thus to increase the accessibility of RNA polymerases to this promoter, activating the transcription initiation.
