pToolkit

pToolkit is a derivative of the λ RED recombinase plasmid pKD46. The heat sensitive origin of replication oriR101 & repA101-ts is replaced by a R6K origin, which is regulated by π protein expressed from the pir gene (BBa_K524004)

Usage and Biology

pToolkit can only be retained in bacterial strains that contain the pir gene (pir+). Recombination through λ RED system can be induced by adding arabinose to the medium.
