The proUP3 promoter appeared to function, given the data in tube A above. However, contrary to expectations, it did not function in the presence of the osmotic inducer as shown in the data for tube B. This was likely due to the high concentration of NaCl supplied in addition to the LB, which already had a relatively high saline concentration. Further experimentation observing the promoter in the presence of varying concentrations of NaCl could be useful to understand more about this functioning promoter.

Applications of BBa_J100032

ProUP3.png

Tube A: E. coli + plasmids containing proUP3 (experimental)
Tube B: E. coli + plasmids containing proUP3 + 20µL of 500mM NaCl (experimental #1 + inducer)
Tube C: E. coli + plasmids containing the placeholder J100028 (negative control)
Tube D: E. coli + plasmids containing pTet (positive control)
Tube E: E. coli + plasmids containing pLux + 20µL HSL-3OC6 (experimental #2 + inducer)
Tube F: E. coli + plasmids containing pLux (experimental #2)
