I noticed that the design of the part incorporates a frame shift mutation, likely causing loss of function of the protein. To fix, either at a C between bases 14 and 15, this will make the part compatible with BioBytes 2.0, or incorporate a T between bases 13 and 14 will reconstitute the wildtype sequence as per the Geneback accession number: EU024648. | |

Applications of BBa_K349119
Digestion at the BsaI cut sites inside of the prefix and suffix will create an A overhang at the 5' end and a B overhang at the 3' end. These ends are compatible with the BioBytes 2.0 assembly method.
