Design Notes

We wanted to only synthesize the promoter with the site of regulation by csgD. After analyzing the sequence, we decided to begin our promoter 20bp before the csgD box, and to finish it on the +1bp of transcription. It corresponds to 71 bp.
We added a non-CDS (non coding sequence) prefix and suffix to our sequence, to fit with iGEM standards.
Need the part BBa_K342003 to be functional.

Source

Our Curli promoter contains the CsgD box of the CsgBA promoter (see reference below for CsgD box discovery) of the strain PHL818 (MG1655 ompR234, see reference below for information about this strain). To design this sequence, we worked on an alignment of different csgA-csgD intergenic regions from various clinical isolated strains with csgA-csgA intergenic regions from PHL818 as a reference. This alignment work has been realized by UMR 5122 (unpublished works, corresponding author philippe.lejeune@insa-lyon.fr).
