Digestion at the BsaI cut sites inside of the prefix and suffix will create an A overhang at the 5' end and a B overhang at the 3' end. These ends are necessary for use in the BioBytes 2.0 assembly method.

Digestion with BsaI around the recognition sites inside of the prefix and suffix will create a kan cassette with an A overhang at the 5' end and a B overhang at the 3' end. These ends are consistent with the BioBytes 2.0 assembly method. This is a base plasmid, meaning that the kan cassette could be digested out with BsaI and replaced with a PCR product that has had BsaI recognition sites and the appropriate AB ends incorporated into it. This allows for the production of more novel AB Bytes.
