Applications of BBa_K320004
This BioBrick has two main fonction, it makes the bacteria to resistant to the Antibiotic Kanamycin and it provides an origin of replication working for E.Coli and Asaia.
To provide the experiment we put the BioBrick in the pSBC3 vector. This vector provide a resistance to Chloramphenicole and an origin of replication which works only for E.Coli.
We transfected E.Coli and Asaia with the plasmid.
The transfect E.Coli were grown on a plat of LB with Agar an containing Kanamycin with a concentration of . This experiment permit us to confirm the providing of the resitance to Kanamycin to teh bacterias.
The transfect Asaia were grown on a plat of Gly with Agar an containing Kanamycin with a concentration of ...(according to our [http://2010.igem.org/Team:EPF_Lausanne/Project_asaia "characterisation of Asaia"]. This experiment permit us to confirm the providing of the origin of replication.
For each strand we made a negativ control with E.Coli or Asaia without plamsid.
We observe that on both plates with transfected bacteria we obtain colonnies and that in negativ controls we obtain non colonnies.

In conclusion we assume that our resitance to Kanamycin works and that the origin in replication also works for Asaia.
