To demonstrate that this biobirck, “FLAG-tag encoding sequence flanked by standard BioBrick™ prefix and suffix”, works as expected, a construct FLAGtag – EGFP was built and ligated into mammalian expression vector pUHD-P3. HtTA1 cells were transfected with this plasmid and observed under florescence microscope 48 hours after transfection. The cells were then lysed and protein extracts obtained. The extracted proteins were then subjected to SDS-PAGE and blotted with M2 (Sigma, anti-flagtag). HtTA1 cells transfected with empty pUHD-P3 were used as control.
