We introduced PPK1 into Shewanella through a codon optimization method and found that it could work normally, and the introduction of PPK1 into Shewanella could also change the efficiency of electricity production by changing phosphorus metabolism. We have also demonstrated that it is also capable of converting Pi to PolyP in Shewanella, and significantly affects the power production of Shewanella, as shown in the figure below.

Figure 1: Electricity production capacity of S. oneidensis. after the introduction of PPK1 We also used transmission electron microscopy (TEM) and scanning electron microscopy (SEM) to illustrate that if the protein is expressed in large quantities in Shewanella, it will lead to excessive intracellular phosphorus content, which will lead to changes in cell morphology.
Figure 2: Scanning electron microscope of the S. oneidensis. morphology of WT and modified S. oneidensis. strains with Strong RBS BBa-B0034

Figure 3: Transmission electron microscopy of the S. oneidensis. morphology of WT and modified S. oneidensis. strains with Strong RBS(BBa-b0034)

When the element was expressed in large quantities, the number of Shewanella decreased significantly, indicating that the mortality rate of Shewanella was significantly increased.
