BBa_K5526001 - Plldr New Promoter Documentation

New Basic Part: BBa_K5526001 (Plldr New Promoter)

Profile

Name: Plldr New Promoter

Base Pairs: 390bp

Origin: Bacteria, synthesized

Properties: The Plldr new promoter, derived from Lactobacillus plantarum, is a lactate-inducible promoter known for its ability to respond to the presence of lactic acid.

Usage and Biology

The Plldr new promoter, derived from Lactobacillus plantarum, is a lactate-inducible promoter known for its ability to respond to the presence of lactic acid. It is a moderately strong promoter, offering controlled expression levels of target genes. Plldr's activity is influenced by lactate concentration, making it useful for applications where gene expression needs to be tightly regulated in response to metabolic changes.

Plldr(new) is a lactic acid promoter that is activated under high lactic acid concentrations, a trait commonly found in tumor areas. It includes several improvements to the original promoter, making it more accurate and resistant to inhibition under low oxygen concentrations.

Cultivation

Plldr(new) is activated by high lactic acid concentrations, a condition often found in tumor environments. The promoter has undergone modifications to ensure it remains active even in low oxygen conditions. Plactate 2 (which includes the Plldr new promoter) plasmids were used as a template for PCR amplification, resulting in a fragment of 3800bp. Figure 2 shows a band consistent with the target size, confirming the successful amplification of the linearized plasmid. After gel recovery, the new Plldr fragment was obtained.
