+ | The HIPPO pathway is a key regulatory pathway in tumor development, with TEAD4 (TEA Domain Family Member 4) being a core transcription factor that promotes the growth and spread of CRC cancer cells [1]. si-TEAD4-1 sequence are designed to target to the untranslated region (UTR) of TEAD4. When siRNA reach the cell cytoplasm, it could binds with the RNA-induced silencing complex (RISC) [2]. Binding with the RISC leads to degradation of the sense strand, whereas the antisense strand continues to be incorporated in the RISC [3]. The RISC then makes multiple mRNA cleavages to downregulate TEAD4 gene expression.

+ | We designed the siRNA against TEAD4 by a special database (https://www.invivogen.com/sirnawizard/). After we input the TEAD4 gene name and sequence, the database would output all the possible siRNA sequence (Figure 1). We chose two pairs of siRNAs with the least off-target possibility.
