Introduction
This composite part is designed to assess whether pRAP can function in cyanobacteria. It includes stayGold, stem-loop, Twister P1, and smURFP. We connect this composite part into a self-replicating shuttle vector that can replicate and function in both E. coli and cyanobacteria[1]. When both green and far-red fluorescence can be detected, we conclude that pRAP is functional in cyanobacteria. This provides a convenient expression system for genetic modification in cyanobacteria, which itself fluorescence in red.
