Linkers can be categorized into three types based on their structures: flexible linkers, rigid linkers, and in vivo cleavable linkers. Flexible and rigid linkers serve the purpose of connecting functional domains, while in vivo cleavable linkers facilitate the release of free functional domains within the body. Apart from their linking function, linkers offer additional advantages in the production of fusion proteins, such as enhancing biological activity, increasing expression yield, and achieving favorable pharmacokinetic profiles.
In recent research by Meyers, W. M., et al (2023), the researcher used 3xGGGGs as a linker in their mammalian cell virus design. They used HEK embryonic kidney cells to test.
3xGGGGs is a commonly used flexible linker for fusing two proteins.
