BBa_J36846 consists of a signal peptide of lipoprotein (lpp), five transmembrane domains of outer membrane protein A (ompA), and streptavidin, wild-type with His6 tag. It's designed to express streptavidin on the cell surface. This part is an improved version of BBa_J36846. BBa_J36846 consists of a signal peptide of lipoprotein (lpp), five transmembrane domains of outer membrane protein A (ompA), and streptavidin, wild-type with His6 tag. It's designed to express streptavidin on the cell surface. To test if the protein successfully expressed on the outer membrane of bacteria, you might need to perform immunostainning analysis or cell fractionation and western blot.
It's a time-consuming work to perform above experiments. To simplify the the procedure and directly observe the expressed protein, we inserted a eGFP-coding sequence lacking a stop codon immediately upstream of streptavidin region. The resulting BBa_K283005 has been demonstrated work as expected (visit our wiki for further information).
