Lambert iGEM attempted to characterize Spinach to validate the improvements of iSpinach-D5-G30-A32 (BBa_K4245000) and iSpinach (BBa_K3380150). iSpinach is a mutant of Spinach developed by researchers at the University of Strasbourg. Compared to Spinach, iSpinach is less salt-sensitive, has higher thermal stability, and produces more fluorescence (Autour et. al, 2016). iSpinach-D5-G30-A32 is a co-crystallized, re-engineered version of iSpinach developed by researchers at the University of Strasbourg. We created BBa_K4245001, a composite part producing Spinach; BBa_K4245002, a composite part producing iSpinach; and BBa_K4245003, a composite part producing iSpinach-D5-G30-A32. The only difference is the basic aptamer part, allowing us to accurately compare the aptamers. However, after failed attempts at cloning, Lambert iGEM decided to continue the characterization of the various Spinach aptamers in 2023.

Lambert_GA 2022

Lambert iGEM attempted to characterize Spinach to validate the improvements of iSpinach-D5-G30-A32 No part name specified with partinfo tag. and iSpinach No part name specified with partinfo tag. . iSpinach is a mutant of Spinach developed by researchers at the University of Strasbourg. We created No part name specified with partinfo tag., a composite part producing Spinach; No part name specified with partinfo tag. , a composite part producing iSpinach; and No part name specified with partinfo tag. , a composite part producing iSpinach-D5-G30-A32. The only difference is the basic aptamer part, allowing us to accurately compare the aptamers. However, after failed attempts at cloning, Lambert iGEM decided to continue the characterization of the various Spinach aptamers in 2023.
