Wild type lac operator is replaced with a perfect palindromic "oid" operator sequence which increases the repression by LacI more than 2.5 fold. To repress the promoter LacI needs to be expressed. Addition of IPTG will derepress LacI activity.
This biobrick is an enhancement of Biobrick BBa_K090501 which has the sequence from pmutin2. BBa_K143015 and BBa_I746665 biobricks have also the wild type lac operator.
