We transformed E.coli TOP10 cells by mistake with this plasmid, and found perfect red cells after over night growth. The restriction of the plasmid with EcoRI and PstI fast digest enzyme showed the expected fragment sizes. We did not continue work with the part, because it was not part of our cloning and assembling plan. Nice colour anyway!.

We've used this part as an alternative to the suicide part p1010 (ccdB) for selecting against cells that have been transformed with a [http://ginkgobioworks.com/support/BioBrick_Assembly_Manual.pdf destination plasmid]. It worked perfectly, and if i might say, it looks smashingly in LB medium ;).
