In order to find an appropriate expression intensity to achieve balance between metabolic burden and detection efficiency, we tried low intensity promoter at first. This composite part contains the low intensity promoter (BBa_J23105) and RBS. The composite part can be directly imported into plasmid and express FXR-ddRFPA1 and RXR-ddRFPB1 at the same time.
We conducted a set of CDCA concentration gradient experiments to measure its fluorescence intensity. The data showed that the fluorescence intensity changed very little under the activation of different concentrations of CDCA. Consistently, no significant differences were observed.
