-Low transformation frequency- repeat transformations were required. Ambiguous bands seen in digest (for both undigested and digested with EcoRI and SpeI). PCR analysis was performed, after the addition of BBa_I0462 to the downstream part of BBa_K081008; in all colonies tested results obtained were not as expected.
