4-1BB is a costimulatory receptor on activated T cells. The ligation of 4-1BB will trigger a signal cascade, which leads to activation of anti-apoptotic molecules, increased cytokine secretion, and enhanced cytotoxicity (Chester et al., 2018). It has been widely used in the second-generation CAR and is found to improve the persistence of CAR-T cells by enhancing CAR-T survival and expansion through non-canonical NFκB signaling pathway (Philipson et al., 2020). In our project, 4-1BB is used in anti-CD19 CAR (<partinfo>BBa_K4175012</partinfo>).

4-1BB

4-1BB is a costimulatory receptor on activated T cells. The ligation of 4-1BB will trigger a signal cascade, which leads to activation of anti-apoptotic molecules, increased cytokine secretion, and enhanced cytotoxicity (Chester et al., 2018). It has been widely used in the second-generation CAR and is found to improve the persistence of CAR-T cells by enhancing CAR-T survival and expansion through non-canonical NFκB signaling pathway (Philipson et al., 2020). In our project, 4-1BB is used in anti-CD19 CAR (BBa_K4175012).
