The C-terminus of PETase is linked to the N-terminus of MHETase using a 12 amino acid serine-glycine linker. Additionally, the part coding for PETase is genetically modified and optimized for PET digestion. The gene is modified such that the 245th amino acid coded is changed from serine to isoleucine. The part has a C-terminus 6xHis-tag for purification.
