Formaldehyde promoter-Inverter-MazF-terminator
It can express toxin protein according to the concentration of formaldehyde.It ensures that our engineering bacteria die when the concentration of glyphosate is low to a certain level.
Usage and Biology
Formaldehyde promoter provides an inducible scheme for downstream genes and proteins. The inverter plays the role of gene inversion which reverses expression pattern when existing. MazF was designed after formaldehyde promoter and inverter, made sense of our kill switch system in degradation: formaldehyde promoter can be activated after formaldehyde induction and MazF expression is blocked and reversed by inverter. In contrast, without formaldehyde, MazF can express under the pl promoter inside inverter and cause cell death by protein toxicity.
