It's noteworthy that, in our Protein Crosslinking design, we intended to adopt spider fibroin as the structural protein modification in order to enhance the tenacity of our artificial leather. Spider fibroin would be expressed in both P. Pastoris and E. coli, respectively. Therefore, codon optimization must be applied in order to avoid homologous recombination and fit the chassis organism. As a result, we synthesized 4 versions of genes coding the same spider fibroin sequence based on the 2 versions of codon optimization for P. Pastoris and 2 versions for the codon optimization of E. coli. Names for those 4 versions of CDS are documented from Spider fibroin optimized V1 to CBM optimized V4, in which the Spider fibroin optimized V1 and V2 are for P. Pastoris, and Spider fibroin optimized V3-4 are for E. coli.

Spider fibroin optimized V1

This part produces spider fibroin that can strengthen the abrasion resistance of the artificial leather.

Pastoris, and Spider fibroin optimized V3-4 are for E. coli.
