Applications of BBa_K081004
Experiment 1 - performed by Lorenzo Pasotti and Mattia Quattrocelli
This device was contained into pSB1AK3. The reporter protein generator K081012 (B0030-E0040-B1006) was assembled downstream of this device. Then, the composite part K081011 (R0051-B0030-C0062) was assembled upstream of this device using BioBrick Standard Assembly.
We transformed 50 ul of Invitrogen TOP10 with the resulting test part and plated transformed bacteria on LB agar plates with Ampicilline.
15 hours after plate incubation at 37°C, we put the plate on the transluminator. Positive transformants glowed under UV rays.
We also picked up some colonies from the plate using a tip and infected 1 ml of LB + Amp. After 3 hours of incubation (37°C, 220 rpm) we watched 50 ul of bacteria with FITC channel. Results are reported in the following figure.
We can see that our Mux CH0 works as expected, because C0061 actually synthesizes 3OC6-HSL, which activates C0062 and R0062 is turned on.
