A P1003 kanamycin resistance cassette with 5 serine -> amber stop codon (TAG) substitutions: S36Amb, S133Amb, S154Amb, S200Amb, S240Amb. This gives kanamycin resistance when co-transformed with BBa_K2725016. This part demonstrates functionality of the semantic containment system for preventing horizontal gene transfer. This part is an improvement upon BBa_K914009.

Figure 2. Graph showing the probability of a read-through (and therefore expression) of a gene with number of amber STOP codons within the coding sequence of the gene.
