The HexaHistidine sequence (GGGGGGHHHHHHGGGGG) was codon optimized for bacterial expression in E. coli and introduced in between amino acids 43/44 in BBa_K2686001 which is shifted by one amino acid downstream compared to the location in BBa_K192000 where it would be in between amino acids 42/43 (Moon et al., 2014).
