The KatG gene encodes for hydroperoxidase I. It protects aerobic, phosphate-starved cells from oxidative damage. The expression of KatG is activated by the global regulator OxyR. This enzyme is specifically upregulated by oxidative agents, as demonstrated by previous research. It has been shown that the promoter of KatG fused to the lux operon can be strongly induced by hydrogen peroxide in a dose-dependent response.

pKatG

The KatG gene encodes for hydroperoxidase I. It protects aerobic, phosphate-starved cells from oxidative damage. The expression of KatG is activated by the global regulator OxyR. This enzyme is specifically upregulated by oxidative agents, as demonstrated by previous research. It has been shown that the promoter of KatG fused to the lux operon can be strongly induced by hydrogen peroxide in a dose-dependent response.

Usage and Biology

iGEM Leiden 2018 has created a screening system for the detection of new antibiotics. We have tested several stress-activated promoters against a repertoire of known antibiotics (Figure 1)

As can be observed in Figure 1 the KatG promoter is activated by hydrogen peroxide.
