Broad host range regulatory element (strength 5)
This broad host range regulatory element consists of a constitutive promoter and a ribosome binding site which can be used for gene expression across different bacterial species. The part sequence was obtained from Johns et al. (ID 34701) and is originally found in Faecalibacterium prausnitzii.
Usage and Biology
The broad host range regulatory element was characterized by Johns et al. among other regulatory elements obtained through metagenomic mining. The relative transcription and translation rate measured by Johns et al. are given below:
The part was characterized in four E. coli strains, S. oneidensis , and P. putida using mKate2 fluorescent protein. Figure 1 shows fluorescence levels after cells transformed with mKate2 under the control of the broad host range regulatory element were grown in LB for 24 hours.
