Colicin E2 DNase Domain with B0034 RBS

The colicin DNase domain BBa_K1976048 is fusioned to the strong ribosomal binding site RBS BBa_B0034.

Figure 1:Colicin E2 DNase domain based on an MD simulation.

Usage

This part can be used for assembly of generators. The So called miniColicin is an active DNase and In combination with the immunity protein Im2 (part part BBa_K1976026) it serves as a safety mechanism (see part BBa_K1976052).

Characteristics

The DNase fragment of colicin E2 has a molecular weight of 15.2 kDa and consists of 133 residues what conforms to about one third to colicin. We used only one of the three domains of colicin since the metabolic burdon would be significant smaller. The activity tests showed that the so called miniColicin has functional DNase activity.
