I used this part for constitutive expression of tetR. After transforming the device DNA from the 2010 iGEM distribution into DH5a E.coli, I miniprepped the sample, cut with EcoRI and PstI, and ran on a gel. The insert was ~2400bp, and the plasmid was ~3000bp. This is quite a bit more than the ~900bp the part is supposed to be. I decided to instead use Q04400 and add a constitutive promoter in front. I did not test to see whether I739001 still expressed tetR.
