Device used to characterize Ribosome Binding Sites through measurement of RFP expression

Device is based on I2056 and contains a partial biobrick site (Spe1, Not1, Xba1) between R0040 and RFP. To insert a BB promoter for testing, digest the promoter Xba1 and Spe1 and the kit EcoR1 and Xba1. Use .5ul of each restriction enzyme during the ligation to ensure the formation of two mixed sites. This design provides the proper spacing, identical to actual construction conditions.
