FliC E.coli coding sequence

This sequence codes for the Flagellin (FliC) protein of Escherichia coli. This protein froms the filament of the bacterial flagellum.

Flagellin is the main protein which polymerizes to form the filaments of bacterial flagella and is necessary for bacterial movement by swimming. The possession of a flagellum allow bacteria to swim towards food sources and so gives them a selective advantage. [1]

It has been demonstrated that Flagellin has the ability to adsorb precious metals such as Platinum, Gold..

Design summary

Starting from the E.coli flagellin sequence ( http://www.uniprot.org/uniprot/P04949 ) we have designed a part to produce this protein in E. coli chassis. All forbidden restriction sites have been removed. Codons have been optimized for E.coli to allow a maximal transcription level.

Proof of functionality

This sequence has been tested: placed under the control of a strong promoter with a ribosome binding site the sequence has been validated as functional. For further information, go on the following link : BBa_K1951008 design
