It was shown that the periplasmic domain of osmolarity sensor protein EnvZ can be exchanged by ligand binding domains of other receptors without compromising its activity (ref. iGEM Tokyo 2012). For easy fusion protein design, we split EnvZ into two parts, removing the periplasmic ligand binding domain to be replaced by other ligand binding domains of interest. This part is designed to form the transmembrane signal transduction and the cytosolic kinase domains of EnvZ. It follows the EnvZ Head domain (BBa_K1909003) and a periplasmic ligand binding domain (e.g. BBa_K1909004). Upon activation of the ligand binding domain, the signal is thought to be transduced to the cytosolic histidine kinase domain, activating it. Once activated, EnvZ phosphorylates OmpR, which acts as a transcription factor activating expression by binding to the Omp Promoter (BBa_R0082). Activation may be monitored by using fluorescence reporters (BBa_K1909013 and BBa_K1909014).

BBa_K1909005 is a protein internal domain part for the design of EnvZ chimeric receptors. EnvZ is part of the well-studied and commonly used two-component system EnvZ/OmpR in Escherichia coli, offering a simple way of signal transduction.
