Design Notes

The lacZ gene from Lactobacillus acidophilus was introduced into the His locus of L. Lactis genome by temperature sensitive vector, which makes the knocked-in strains become blue on plate with X-Gal (5-bromo-4-chloro-3-indolyl-β-D-galactopyranoside).

Using the genomic DNA of Lactobacillus acidophilus as the template, the lacZ gene product is obtained by PCR and cloned into the pSB1C3 plasmid.
