Cas3 protein with pLuxcI right promoter and weak RBS.
Gene encoding I-F type CRISPR_Cas system Cas3 protein, which is a helicase and ssDNA nuclease. Attached there is a pLux/cI right promoter and weak RBS site. We wanted to have different expression strengths of Cas3, so we cloned it also to a strong RBS (BBa_K1773019), and a medium RBS (BBa_K1773020).
In the I-F type CRISPR-Cas system Cascade complex (BBa_K1773005) distinguishes target DNA, and rectruits Cas3 protein, which acts as a nuclease and helicase, which hidrolises ssDNA in a 3'-5' direction.
