Usage and Biology

Usage:

  • This part was created by iGEM@Imperial 2006 to be used as the prey part in their 2006 project [http://openwetware.org/wiki/IGEM:IMPERIAL/2006 'Engineering a Molecular Predation Oscillator'] to generate a the production of Acyl-Homoserine Lactone (a quorum sensing molecule) using a positive feedback loop.

Biology:

  • The positive feedback loop relies on leaky expression of LuxR by the tetR promoter which causes production of luxR. AHL already present in the cell forms a complex with luxR and lux pR promoter, expressing LuxI and GFP. LuxI enzymatically produces AHL. GFP can be used to monitor transcription of LuxI as a check for the functionality of the part.
