Inducible ampicillin resistance cassette (regulated by RhlR + N-buturyl-HSL)
This device is a combination of BBa_K1073001 and BB_K1073000 and mediates a inducible ampicillin resistance. It includes the inducible promoter of the rhl quorum sensing system of Pseudomonas aeruginosa followed by a RBS and the ampicillin resistance gene coding for beta-lactamase.
Usage and Biology
The promoter is activated in the presence of RhlR in combination with the autoinducer molecule N-buturyl-homoserine lactone produced by RhlI. The expression of the beta-lactamase can therefore be induced and tuned by supplying RhlR and N-buturyl-HSL.
Note that the promoter is leaking to a certain degree when not activated. For further information and experience on this device see BBa_K1073035.
