The plasmid seems to be missing the X and S restriction sites. Digestion by these enzymes fails Agarose electrophoresis of part K206009 as ordered from the registry in spring 2011 digested with several iGEM enzymes. nd stands for "non digested" As shown above, the digestion by P linearizes the plasmid as expected but X and S have no effect compared to the non digested plasmid. Moreover, double digestion by X+P only linearizes the plasmid.
