We believe that this is due to the transcription factor PhoP binding to the <a href"https://parts.igem.org/BBa_K902009> mgtA promtoer</a>. Since there is no mgtA promtoer in this circuit we believe that the response is not dramatic enough, i.e there are not enough transcripts produced. However we still see some supression with increasing concentration of magnesium. This part was built to characterize the mgtA riboswitch. Putting the mgtA riboswitch under the control of the TetR promoter allowed us to test the regulation capabilities of the mgtA riboswitch alone under different concentrations of magnesium. This part was built as a control for the mgtA system. For the experimental protocol click here. This part works as expected.
