This construct was designed with the BBa_R0010 in order to control transcription. Transcription of 1-SST and 1-FFT greatly inhibit bacterial growth with the production of plant inulin. LacI protein and CAP can succesfully inhibit transcription of both genes to allow the bacteria to grow. With the addition of IPTG to the medium, LacI will be inhibited and transcription can resume.

Reporter system for SST and FFT (BBa_K899000 and BBa_K899001)

Reporter system to test for transcription of fructan:fructan fructosyltransferase (FFT) and Sucrose:sucrose fructosyltransferase (SST) genes with green flourescent protein (GFP) as reporter, and lacl as promoter.

This construct was designed with the BBa_R0010 in order to control transcription. Transcription of 1-SST and 1-FFT greatly inhibit bacterial growth with the production of plant inulin. LacI protein and CAP can succesfully inhibit transcription of both genes to allow the bacteria to grow. With the addition of IPTG to the medium, LacI will be inhibited and transcription can resume.
