The part was designed so that LacI transcription/translation does not affect the transcription/translation of CysDes. In other words LacI+lacIq promoter are in the reverse direction. Tac promoter + Lac operator + CysDes are in the forward direction.
Source
LacI+LacIq promoter comes from pET-21b plasmid. CysDes gene comes from the genome of Treponema denticola.
