Design Notes
The yeGFP part (BBa_K319039) from last year didn't have a stop codon so this tagged promoter should have a terminator sequence cloned downstream before using. We defined the distal region as the most upstream 516 nucleotides of the Adh1 promoter and the most downstream 207 nucleotides of the Gal1 promoter.
Source
The distal region of the Adh1 promoter was PCR amplified from a plasmid source of BBa_K319005 and using overlap extension PCR was fused to the amplified proximal region of the Gal1 promoter. The Gal1 promoter is a commonly used promoter in our lab and we amplified it from a plasmid source. yeGFP was PCR amplified from a plasmid source of BBa_K319039.
