pToolkit

pToolkit is a derivative of lambda RED recombinase plasmid pKD46. The heat sensitive origin of replication oriR101 & repA101-ts is replaced by a R6K origin. The plasmid is selected for under incubation of less than 30 degree Celsius with ampicillin.

Usage and Biology

pToolkit can only be retained in bacterial strains that are pir+. Recombination through lambda RED system can be induced by adding arabinose to the medium.
