BBa_K316009 Penn State 2011 iGEM

Part of our reporter project was largely focused on improving the part developed by Imperial College London iGEM 2010 (BBa_K316007) we began by characterizing their test part K316009. The data for this test, using the standard catechol assay described by their team <a href="http://2010.igem.org/Team:Imperial_College_London/Protocol">here</a> is shown below. For induction 10 ul of 100uM of IPTG was used.

This data shows that, while their part is capable of extremely fast conversion of catachol to muconic acid, it is also subject to extreme sensitivity that leads to significant results of color production seen from both induced and un-induced cells.
