Characterisation

We transformed Bba_K091104 (=pLac/Mnt hybrid) in DH5α-cells, followed by a minipreparation. After this we did a restriction digest with EcoRI and SpeI. Since the pSB1A2 vector is 2079bp and the Lac/Mnt hybrid promoter should be 87bp, we also expect a fragment of 87bp, one of 2079 and one of 2166bp. However, we see a fragment of 1000bp and one of ±2000bp.
