This part is a loading sequence-and-replicase-deficient version of the cDNA of phage MS2, BBa_K313006. (XbaI site is removed.)
Virus transformed from this part carry out mRNA with specific loading sequence(19bp)

and infect another E.coli.

This cDNA does not have the loading sequence, BBa_K313010, which is necessary for loading of RNA into the capsid of MS2 phage. So, MS2 phage produced from this part cannot load the RNA of itself. Instead, the capsid is assumed to load any mRNAs which includes the loading sequence.
In the infected E.coli, translation of mRNA occur.

MS2 phage cDNA (loading sequence and replicase-deficient)

This part is a loading sequence-and-replicase-deficient version of the cDNA of phage MS2, BBa_K313006. (XbaI site is removed.)

This cDNA does not have the loading sequence, BBa_K313010, which is necessary for loading of RNA into the capsid of MS2 phage. So, MS2 phage produced from this part cannot load the RNA of itself. Instead, the capsid is assumed to load any mRNAs which includes the loading sequence. This mutant phage will be able to carry mRNAs you want to various cells as information.

As this mutant phage has lost the coding region of its replicase, its arbitrary proliferation will be prevented.
