Ligand
The ligands of DAF-12 are various isomers of dafachronic acids, which derive from sterols upon oxidation by DAF-9, a cytochrome P450 enzime.
Physiological effects of DAF-12
daf-12 in Caenorhabditis elegans plays a key role in the regulation of development and determination of adult longevity. It has effects also on resistance to oxidative and thermal stress which makes daf-12 the first gene with alleles that can extend or shorten lifespan.
Possible applications of GAL4-DAF-12
By the addition of lipid ligands (dafachronic acids), the created system activates the expression of any kind of genes fused to UAS(upstream activation seqence)previously. Also can be used as a lipid sensor by activating reporter genes (e.g. Luciferase).
