This is a photo of the luciferase when luciferin is added to the LB broth the cells were grown in.
This is a photo of the luciferase when the cells are resuspended in citrate buffer, pH4.8. This allows better entry of the luciferin into the cells, but also shifts the wavelength of a proportion of the enzymes towards the red end of the spectrum. The yellow colour is due to a mix of green emitting luciferase, and red emitting luciferase.
