The loxP site is the substrate of the Cre recombinase. In nature the site is a palidromic 34bp sequence with a directional 8bp core. Because the site is not a multiple of 3, if it is used within coding regions it will throw the remainder of the sequence out of frame. We modified the loxP sequence by adding 2 bases to the 3' end. The resulting biobrick can now be used within transcribed assemblies without modifying the frame or the surrounding biobricks.
